Biomolecule Conjugation Protocol
1.Take an appropriate amount of beads, wash several times with conjugation buffer to completely remove isopropanol, then separate magnetically for later use.
2.Disperse the desired biomolecule in carbonate buffer (pH 9.0), resuspend the beads in this solution, and conjugate at 37 °C for 2 hours or at 4 °C overnight.
3.After conjugation, separate the beads magnetically and discard the supernatant. Resuspend the beads in carbonate buffer (pH 9.0) containing 1% BSA, then incubate at 25 °C for 1 hour to block unreacted tosyl groups.
4.After blocking, separate the beads magnetically and discard the supernatant. Wash the beads three times with 20 mM PBS buffer (pH 7.4), resuspend, and store at 4 °C. If needed, add 0.02% sodium azide to inhibit bacterial growth.
